Freestanding Porous Polymer Thin Filmsfor the Study of Cell-Cell Communications Mediated by Cytokines |

Vital cellular functions are greatly influenced by surrounding cells and environmental factors. Particularly, communications between cancer and stromal cells have received intense attention and the analysis on cell-cell interactions has been a growing and significant issue in cancer research. Herein, we have developed novel cell co-culture platforms based on freestanding cellulose acetate (CA) thin films with controllable pore size for more accurate in-vitro analysis on the communications between human breast metastatic cancer cells and three different types of stromal cells. The porous CA freestanding membranes enable us to trace the behavior of metastatic cancer cells co-cultured with stromal cells as well as to analyze cytokine-based paracrine communications as a function of stromal cell types. Furthermore, the effect of the distance between cells and the membrane morphology on the secretion of cytokines has been investigated using the detachable and porous CA membrane platforms. |
