제목 | Development of ratiometric fluorescent peptide-based probes with high binding affinity for Pb(II) |
초록 | Pb(II) contamination is detrimental to human body because Pb(II) accumulation in human body can cause in anemia, renal impairment, or neurodegenerative pathologies. Thus, detection of lead ion in live cells and aquatic environment is crucial for human health. Herein, two fluorescent peptide-based probes (1: Cy-MA-OH, 2: Cy-AhxMA-OH) bearing a cyanostilbene fluorophore were synthesized and studied for the detection of Pb(II) ions. 1 and 2 exhibited ratiometric response upon addition of lead ions in aqueous buffered solution (pH 7.0 HEPES containing 1 % of DMF). Isoemissive point showed at 535 nm and excimer-like emission was to be 575 nm. For both probe 1 and 2 (5 μM), 2 equivalent of lead ions were sufficient to be saturated and Kd value was calculated to be 5.58×10-6 M and 4.62×10-6 M respectively. Furthermore, selectivity and interference study of the probes were carried out. Among 16 different other metal ions, Pb(II) was most selective to the probe 1 and 2. |
