제목 | Self-Assembled Protein Molecules for the Application to Protein Chip |
초록 | Highly oriented antibodies on the solid surfaces have been required to attain the ultimate selectivity as well as sensitivity. Because of the simplicity and adaptability of self-assembled monolayers (SAM) and control over biomolecule surface orientation, SAM can play an important role in the construction of the antibody-based biosensors. In this study, to fabricate a biocompatible surface and endow the orientation of antibody molecules, the Fc receptors of immunoglobulin G (IgG) were introduced on the solid surface. In order to immobilized these binding proteins, surface modifications of protein molecules and substrate were carried out, respectively. The immobilization of the proteins molecules was investigated by surface plasmon resonance (SPR), and the topography of the self-assembled protein molecules on the surface was analyzed through atomic force microscopy (AFM). On the fabricated protein layer, various kinds of IgG can be immobilized, which can be used to construct the immunosensor or protein chip. |
