Synthesis of smooth and continuous DNA nanowires, preserving the original structure of long native DNA and allowing its analysis by scanning electron microscope (SEM), has been demonstrated. Gold nanoparticles densely assembled on the DNA backbone via thiol-tagged DNA binding peptides work as seeds for uniform metallization of DNA templates. Using this approach, we were able to visualize smooth and uniform structures of DNA (48.5 kb), its dimer (97 kb), and trimer (145.5 kb) using the scanning electron microscope. This method also allows whole analysis of micro/millimeter sized DNA molecules with entangled 3-dimensional features. |
