Outgrowth endothelial cells-specific capture by antibody immobilization on nano-structured surface |

Restoration of damaged endothelium might be a well-built way to reduce late stent thrombosis and restenosis. It has been reported that outgrowth endothelial cell (OEC) can differentiate to endothelial cell and then facilitate re-endothelialization. Some groups have developed OECs capture strategy using antibody (Ab) immobilization technologies for the restoration of endothelium. We focused on CD146 as a target surface biomarker for OEC capture. In this study, it is seemed that anti-CD146 Ab-immobilized stent can capture circulating OECs and restore damaged endothelium. Fabrication of silicon nanofilamentwas accomplished for the formation of nano-structured surface. SiNf and anti-CD146 Ab-immobilized surfaces were evaluated by FTIR and contact angle. Anti-CD146 Ab-immobilized surface was evaluated by OEC capture assay. Our results exhibited that anti-CD146 Ab-immobilized surface captured selectively and significantly more OECs than bare surface and anti-CD34 Ab-immobilized surface. |
