A Reduction and pH dual-responsive biodegradable polypeptide micelles for effective treatment of liver cancer |

A series of pH and reduction biodegradable polypeptide micelles were developed based on disulfide-linked methoxy poly(ethylene glycol) -b-poly(benzyl-L-glutamate) -co-poly[2-(dibutylamino)ethyl-L-glutamate](mPEG-SS-PDAELG) copolymer and applied to act as efficient intelligent carriers to rapid trigger intracellular release of doxorubicin (DOX). mPEG-SS-PDAELG was synthesized by a combination of ring-opening and aminolysis reaction. The pH/reduction-responsive behavior of mPEG-SS-PDAELG was investigated by both dynamic light scattering and fluorescence microscopy. The micelles morphology was confirmed by TEM. The in vitro release studies showed that the DOX-loaded micelles showed retarded drug release at tumor extracellular pH but accelerated drug release at weakly acidic or reductive environment. Our findings suggest that pH/reduction-responsive DOX-loaded micelles selected as a model drug to effective treatment liver cancer. |
