Intracellular Delivery of siRNAs, Plasmids and Proteins to Hard-to-Transfect Cells Using Carbon Nanosyringe Arrays |

In spite of advances in commercial transfection reagents and common methods, there are still a number of cell lines and primary cells that are hard-to-transfect cells and problems remained unsolved. To resolve these issues, we report a highly efficient, less toxic and optimized form of carbon nanosyringe arrays (CNSAs) that are capable of delivering various biological cargo into immortalized and primary mammalian cells. Major advantages of the CNSAs are that it is able to penetrate the cell membrane with native form of biological cargoes. We have shown that siRNAs, plasmids and proteins can be efficiently delivered to diverse cells including primary mouse lymphocytes. These CNSAs may prove to be novel yet simple transfection method that could be used for efficient intracellular delivery of various cargo even to cells that are hard to transfect. This serves as a new pathway for studying cells that are previously impossible due to lack of methods for successful gene transfer. |
