Thermo-responsive nanoparticle-mediated delivery of retinoic acid into human induced pluripotent stem cells for enhanced neuronal differentiation |

Neuronal differentiation of human induced pluripotent stem cells (hiPSCs) has been studied for application to diverse neurodegenerative diseases. Retinoic acid (RA) is a transcriptional factor known to enhance neuronal differentiation of neural stem cells or progenitor cells. In this study, we report thermo-responsive poly(N-isopropylacrylamide-co-acrylamide) (PNIPAM-co-Am) nanoparticles to efficiently deliver RA into hiPSCs for neuronal differentiation. The synthesized PNIPAM-co-Am nanoparticles exhibited the change of nanoparticle size depending on incubation temperature. The PNIPAM-co-AM nanoparticles incorporated with RA were efficiently transferred into hiPSCs and significantly promoted neuronal differentiation of hiPSCs. In conclusion, thermos-responsive nanoparticles developed in this study could be useful for improving therapeutic efficacy of hiPSCs.
