In recent years, a cell spheroid system is considered as most common way to culture cells in 3D. However, limited size, structural stability and heterogeneous cell differentiation due to the insufficient supply of oxygen/nutrients into the central region are still remained as challenges. In this study, we fabricated porous leaf-stacked structure particles throughout entire matrix (LSS) which can provide space for sufficient supply of oxygen/nutrients, act as improved structural stability, and allow sustained release of signaling molecules for induction of differentiation throughout whole cell spheroids. A cell spheroid system was prepared using the LSS particles and human bone marrow-derived mesenchymal stem cells (hBMSCs). The morphology, release pattern of signaling molecules from LSS particles, cell survival/structural stability/osteogenic differentiation of hBMSCs/LSS particle spheroid were observed. We also evaluated hBMSCs/LSS spheroid for promoting bone regeneration in vivo. |
