제목 Synthesis and micellar characterization of triblock copolymers by using methoxy poly(ethylene glycol)-block-poly(caprolactone) diblock copolymers
초록 Micelles composed of amphiphilic block copolymers have been shown to possess significant potential as delivery systems providing several advantages when used as a drug delivery system for hydrophobic drugs. Typically, copolymer micelles of this type are less than 100 nm in diameter, leading to prolonged circulation and altered biodistribution of drug encapsulated within micelles. Recently, amphiphilic block copolymers, in particular poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) or Pluronics®, have been shown to chemosensitize drug-resistant cancer cell lines. In addition, the molecular structure, shape, and hydrophilicity of the polymer have great effects on the micelle properties.
Diblock copolymers of methoxy poly(ethylene glycol)-block-poly(caprolactone) was synthesized and characterized in order to assess the potential of these copolymers as a micellar drug-delivery system. Synthesized diblock copolymers was a MePEG molecular weight of 2000 g/mol and PCL molecular weight of 900 g/mol. Accdrding to using MPEG-PCL diblock copolymer, we synthesized triblock copolymer, MPEG-PCL-PCL, MPEG-PCL-PLLA, and MPEG-PCL-PLGA by the ring-opening polymerization of using the hydroxyl group of MePEG as the initiator. Molecular weight of terminal type was 900 g/mol. The synthesized tiblock copolymers were characterized with 1H NMR, GPC, DSC, and XRD. The micellar characterization of MPEG-Polyester tiblock copolymers in an aqueous phase was carried out by using NMR, atomic force microscope(AFM), and fluorescence techniques. As the degree of polymerization of terminal types, we acquired critical micelle concentration (CMC) values and type and diameters of micellar core.
This work has shown that triblock copolymers could be successfully synthesized using a ring opening polymerization. We suggest that these copolymers may be suitable micellar carriers of hydrophobic drug.
