Expression of Recombinant Protein with Unnatural Amino Acid via Suppression of AGGA Four-base Codon |

Adding new amino acids into the set of building blocks for protein synthesis expands the scope of protein engineering. Orthogonal pairs of tRNA and aminoacyl tRNA synthetase have been developed to incorporate unnatural amino acids (UAAs) into codons such as stop codons and four-base codons that are generally not used in organisms for coding amino acids. In particular, the tyrosyl-tRNA and tyrosyl-tRNA synthetase pair from Methanococcus jannaschii has been engineered for incorporating diverse UAAs into the amber codon. We were interested in taking advantage of the system to incorporate UAAs into four-base codon such as AGGA because the Mj tyrosyl-tRNA synthetase had been reported to have promiscuity toward anticodon. In this presentation, we show that the engineered Mj tyrosyl-tRNA synthetases activated MjtRNAUCCU without any additional engineering toward the tRNA anticodon and were used to incorporate UAAs quantitatively in response to the AGGA codon. |
