Biocompatible Bimodal Imaging Contrast Agent using Gold Nanoparticle |

Gold nanoparticles (AuNPs) have attracted much attention as the potential agent for in vivo therapeutics and diagnostics (theragnosis). However, AuNPs cannot be used in the biomedical field because of the instability in physiological condition. To improve stability of the AuNPs in physiological condition, AuNPs were coated with biocompatible Glycol Chitosan (GC). AuNPs were characterized by zeta-potential, dynamic light scattering, HR-TEM, stability test in PBS. GC Coated AuNPs (GC-AuNP) were intravenously tested as a Computed Tomography (CT) contrast agent, in vivo mice tumor (SCC-7) model. Because the AuNPs have a quenching effect, the AuNPs were conjugated with Cy5.5-MMP (Matrix MetalloProtease) specific substrate, which can be used as a MMP specific fluorescence imaging probe for tumor progression. The Cy5.5-MMP conjugated GC-AuNPs were intravenously injected in mice bearing SCC-7 tumors for tumor detection. The GC-AuNPs showed their accumulation in tumor by EPR effect. |
