Design of Water Soluble Paclitaxel Based on Hyperbranched Polyglycidols |

Current clinical formulation such as paclitaxel (PTXL) contains a 50:50 mixture of Cremophore EL and ethanol to enhance the solubility of PTXL in water. However, a large number of studies have reported various side effects of Cremophore EL such as hypersensitivity, neurotoxicity, and neuropathy. To overcome the serious side effects, chemical conjugates of PTXL and hyperbranched polyglycidols (HBP) were synthesized by utilizing glutaric anhydride as a “bridge” with esterification in dimethyl formamide. A series of HBP-PTXL conjugates (HPCs) with different molecular weight of HBP and different ratio of HBP to PTXL were synthesized and characterized. An intact form of PTXL was regenerated from HPCs at acidic or basic pH condition. Water soluble HPCs exhibit more enhanced cytotoxic effect for cancer cells suggesting that they can be potentially utilized as tumor-specific injection therapeutic agents. |
