제목 Efficient production of gamma-aminobutyric acid by recombinant Escherichia coli
초록 Inthis study, the effect of gene knockdown of sucA, which encodes enzyme of succinylCoA synthetase, on GABA production was investigated in recombinant Escherichiacoli. For this, using the CRISPR interference system, E. coli GTSstrain, in which sucA gene was knocked down, was prepared from the E. coli GB200 strain whose GABA productionactivity was previously confirmed. From flask cultures performed under neutralpH condition, it was confirmed that the E. coli GTS strain produced GABA2.18-fold higher than the parent strain. For the development of mass productionprocess of GABA, fed-batch cultures of E. coli GTS were carried out in a7L-fermenter by varying the variables such as inoculum size and glucose feedingrate. As a result, E. coli GTS was observed to produce up to 50.3 g/L GABA by consuming glucose510 g in 59 h, indicating that the E. coli GTS strain has an outstandingpotential for GABA production.
