Thermosensitive Hyaluronic Acid/Pluronic F127/Microsphere Composite Hydrogel for Cartilage Tissue Engineering |

Three-dimensional hydrogel is a useful cell carrier for injectable implantation. Hyaluronic acid (HA) is an important component of the extracellular matrix (ECM) of connective tissues. F127 Hydrogel which displays temperature responsiveness and non-toxicity is a good candidate for drug-delivery systems. In this study, we hypothesize that HA/F127/microsphere composite hydrogel may have some advantages in the cartilage formation. For in vitro and in vivo tests, chondrocytes were encapsulated in the hydrogel, mixed with microspheres. The cell-incorporated hydrogels were cultured in 24-well plates for two weeks and then implanted into nude mouse subcutaneously. The composite constructs were evaluated using Safranin O staining for GAG. Cartilage-specific gene expression was also determined using RT-PCR. This composite hydrogel system is expected to be useful as an injectable hydrogel for cartilage regeneration. |
