Reduction of Inflammatory Reaction in Purified Alginate Microcapsules |

Alginate, an anionic polysaccharide extracted from brown seaweed, remains the most widely used biomaterial for immobilizing cells to be transplanted. However, the main problem is the immune rejection in vivo. To overcome this drawback, we have developed using modified Korbutt’s method for alginate purification. After alginate microcapsules manufactured, NIH/3T3 fibroblast and RAW 264.7 macrophage cells were seeded in microcapsules. Cell viability was analyzed by MTT assay. RT-PCR was performed to assess mRNA expression for inflammation cytokines such as TNF-α. Alginate microcapsules were implanted into a wister rat, and the implanted alginate microcapsules were extracted after 1 and 2 weeks. Tissues surrounding implants were harvested and evaluated by histology through H&E and immunohistochemistry through ED-1 staining. This study showed that purified alginate reduced inflammation reaction in vitro and in vivo. This research was supported by WCU (R31-20029) and XRC (A040004). |
