Even though lipids are known to be penetration enhancers, intact liposomes do notact like lipid molecules because those lipids in liposomes form well-assembled bilayers,which can hardly penetrate into the stratum corneum. If intact liposomes cannotpermeate through the skin membrane and drug particles are embedded in the bilayers,the drug particles would have fewer chances of penetrating into the skin membrane, andthe overall drug diffusion rate will be decreased.In this study, the effects of liposomes in the diffusion of drug molecules through the pigskin were investigated. Two hydrogel samples, one containing liposomally encapsulatedhydrocortisone acetate and the other containing free-drug HCA, were tested for drugpermeation through the pig ear skin mounted on diffusion cells and also for drug releasefrom the hydrogel in a similar environment. It was found that liposomes impeded drugdiffusion through the skin while the mass fluxes of drug molecules from bothliposome-hydrogel and free-drug hydrogel were almost the same.
