As skin keratinocytes are frequently exposed to radiation, biologicalresponse of human keratinocytes to various radiation doses and time-points is important.Here, we applied multiomics approaches to identify the biological effectsdepending on IR dose and time in HEKn cells on Day 0 and 2 after irradiationwith 0, 0.1, 0.5, and 2 Gy. The expression levels of proteins and metabolitesinvolved in DNA repair system, energy metabolism, and epidermal cornificationwere changed after irradiation. On Day 2, fumarate and fumarate hydratase,which promote DNA repair, were increased in response to 2 Gy and all doses inducedthe proteomic changes correlated with epidermal cornification and inhibition ofcell death. Also, we observed dose-dependent increases in the cornified layerand the expression of involucrin using a human skin 3D full-thickness model. Takentogether, we provided comprehensive insight to biological effects of IRapplications on human epidermis. |
