An industrially important platform chemicals, 3-hydroxypropionic acid (3-HP) and malonic acid (MA) was successfully produced through the β-alanine (BA) route by metabolically engineered Escherichia coli. First, to produce 3-HP and MA from BA, various combinations of downstream enzymes were screened and BA pyruvate transaminase from P. aeruginosa was selected. This platform strain was further engineered by introducing E. coli MSA reductase to reduce MSA to 3-HP. To increase its production, native promoter of the sdhC gene was replaced with the strong trc promoter. To produce MA, E. coli semialdehyde dehydrogenase was introduced to oxidize MSA to MA and additionally, ydfG gene was deleted. Fed-batch cultures of these final engineered strains resulted in production of 31.1 g/L 3-HP or 3.60 g/L MA from glucose.
