Structure modification of water-soluble conjugated polymers for enhanced sensitivity of FRET-based DNA detection |

Water-soluble cationic conjugated polymers (CCPs) with two different counterions are studied and compared as a donor in fluorescence resonance energy transfer (FRET) to fluorescein-labeled single-stranded DNA (ssDNA-Fl). Fluorescence quenching of the acceptor (Fl) via photo-induced charge transfer (PCT) in the polymer/ssDNA-Fl electrostatic complex should be minimized to improve DNA detection sensitivity. The energy-wasting PCT can be controlled by simply exchanging counterions accompanied for charge compensation of the CCPs. Large counterions perturb the D-A intermolecular distance in molecular scale, resulting in a substantial decrease of PCT and enhanced FRET-induced Fl signal. Structure modification of CCPs using different counterions can fine-tune the competition between energy transfer and PCT quenching for optimized FRET-based DNA sequence detection. |
