Quantitative-Analysis of Pulp-Derived Soluble Oligosaccharides by an Enzymatic-HPLC Method
A simple, rapid enzymatic-HPLC method was devised for the quantitative analysis of pulp-derived soluble oligosaccharides. Cello-, xylo-, and manno-oligomers were completely hydrolyzed by a crude enzyme mixture (Acucelase) of cellulases and hemicellulases from Aspergillus aculeatus no. F-50 for 1.0 h at 50 degrees C. After the Acucelase treatment, the neutral sugars formed were separated and quantified by high-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD). This method was successfully applied to the quantitative analysis of oligomers formed by the enzymatic hydrolysis of pulp.
