Natural killer (NK) cells are a special immune effector cells to directly attack cancer cells. We designed multifunctional polymeric biomaterials for an individual NK cell coating to enhance its anticancer efficacy. This ex vivo engineering materials contain 6 functional moieties; cell surface anchoring lipid, PEG intracellular penetration blocker, positive charge moiety, fluorescence dye, cancer recognition moiety, and stimuli responsive degradable moiety. Various intrinsic properties (i.e., proliferation, cytokine production, availability of membrane receptors/ligands) of coated NK cells were maintained. Cancer recognition of NK cells were effectively enhanced by folate-mediated binding. Subsequently, anti-folate was released via cleavage of disulfide bones in dissociated coating material in response to glutathione leaked from disrupted cancer cells, and induced secondary anticancer efficacy. Hence, our multifunctional NK cell coating could be utilized as an efficient immunotherapy. |
