Fabrication of Gold Nanoshell-formed Nanoparticles Containing Porphrins for Combined Photoherapy of Cancer |

Photo-active nanoparticles for cancer therapy have been used for photothermal and photodynamic therapy. Photodynamic therapy(PDT) is excellent method for the treatment of cancer. It involves the systemic administration of photosensitizers(PS) like porphyrin, so PS transfers photon energy to generate cytotoxic reactive oxygen species(ROS) to kill cancer. In the case of photothermal therapy(PTT), it involves the conversion of absorbed photon energy into the heat by using gold nanomaterials. In this work, 3rd generation dendrimer porphyrin with 32 carboxyl groups was deposited onto gold nanoshell coated silica nanoparticles via electrostatic interaction. In order to confirm photodynamic and photothemal effect of nanoparticles under light exposure, ROS generation and temperature increase test were conducted. Furthermore, synergetic combined phototherapy including PDT and PTT was achieved through cell viability using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. |
