Stem cells with the capacity to self–renew and to differentiate into specific cell types have been considered as a promising cell source for regenerative medicine, tissue engineering and disease modeling. However, development of cell culture system for reliable, reproducible, scalable production of the stem cells is remained as a challenge for biologists and material scientists. In recent, a variety of cell culture systems based on embryonic fibroblast feeder, matrigel, hyaluronic acid are adapted for stem cell proliferation, and encouraging outcomes are reported. However, the contamination by animal-derived cell, animal pathogen transmission, difficult cell harvesting are considered as drawback for clinical applications. In this study, we developed a 3D cell culture system consisted of intact hyaluronic acid and membrane with selective permeability. The morphology/permeability of membrane, and cell proliferation behavior in 3D culture system were investigated. |
