The robust nanocarriers were developed by calcium phosphate mineralization on PEG-b-polyaspartate-b-polyphenylalanine polymer micelles for effective anticancer drug delivery. Cy5.5, a NIRF dye, was conjugated with polymer micelles to monitor their in vivo biodistibution. The mineralized layer could minimize the loss of the entrapped doxorubicin (DOX) before reaching target tissues, and selectively dissolve in lower pH and Ca2+ concentration of endosomal compartments to facilitate intracellular drug release. In vivo tissue distribution and tumor accumulation of hybrid nanocarriers labeled with the near-infrared (NIR) fluorophore, Cy5.5, were monitored in MDA-MB231 tumor-bearing mice. To assess in vivo antitumor efficacy, we performed the tumor volume monitoring for tumor-bearing mice. The growth of tumors was remarkably inhibited, as compared with control groups, due to the stability of mineralized micelles and enhanced permeation and retention (EPR) effect. |
