제목 Enhanced Gene Delivery effects of PAMAM dendrimer by Surface Modified with NLS from Fibroblast Growth Factor 3
초록 Polyethyleneimine(PEI) polyamidoamine(PAMAM) dendrimer and poly(L-lysine) were contained between favor cationic polymers. Particularly, PAMAM dendrimer is a cationic polymer used in the gene delivery system study. It has special characteristics such as equal size distribution, low cytotoxicity and relatively good transfection efficiency but it shows less transfection capacity in comparison with PEI. To improve the transfection efficiency of PAMAM dendrimer, we modified it with a nuclear localization signal(NLS) peptide. NLS is a specific peptide facilitating translocation into the nuclear region. In addition, a mouse fibroblast growth factor 3 protein contains a bipartite NLS motif. The entire sequence of FGF3 is RLRRDAGGRGGVYEHLGGAPRRRK. It has two functional sequences of RLRR and RRRK at N-ter and C-ter. In this study, we introduced the RLRR segment peptide sequences on the surface of PAMAM dendrimer and characterized its gene carrier activity using HEK293, NIH3T3, HeLa cell lines.
