Fabrication and characterization of PLGA polymer-lipid hybrid nanoparticle for gene delivery platform |

The major problem in the field of cell therapy is how to effectively deliver target genes to special cells. In this study, we aimed to develop a poly (D, L-lactide-co-glycolide) (PLGA)-lipid hybrid nanoparticles (NP) for gene delivery, which has the ability to overcome the disadvantages of polyethylenimine (PEI) - or cationic liposome based gene carrier. The PLGA polymer-lipid NP were successfully fabricated by emulsion solvent evaporation method and characterized by scanning electron microscopy, dynamic light scattering. Plasmid EGFP gene as a model gene was complexed with NP (100-200nm) and then transfected to 293 cells. Therefore, PLGA polymer-lipid hybrid nanoparticles would be widely utilized as a promising non-viral carrier for the delivery of genes as well as drugs. |
