The encapsulation of proteins using PLGA nanoparticle has been investigated for delivery of pharmaceutical proteins. However, protein instability has been a major hurdle during primary emulsification process due to generation of w/o interface, shear stress and heat. In this study, the effect of various stabilizing additives on 3D8 in primary emulsification process in the presence of PLGA was studied. 3D8 was obtained as a monoclonal antibody having anti-viral activity. We produced 3D8 loaded PLGA nanoparticle by w/o/w double emulsion method. 3D8 was successfully recovered up to 90-100% by adding of PEG 600 to w/o emulsion containing PLGA. The secondary and tertiary structure of 3D8 was studied by CD and fluorescence spectroscopy. We also performed ELISA and DNA hydrolyzing assay to investigate catalytic activity of 3D8, respectively. As a result, we suggest that mannitol and heparin are suitable stabilizers to maintain biological activity as well as structural stability of 3D8. |
