In this study, optimization of gamma irradiation on degradation of kenaf core by alkaline pretreatment was investigated. The kenaf core was carried out at doses of 0 (control), 200, 500, 700 and 1,000 kGy using a Co-60 gamma radiation source. The irradiated sample was treated by alkaline solution (3% sodium hydroxide, v/v) using an autoclave for 5 h at 120 oC. And then, we were obtained sugar yield that is amount of produced monosaccharide in pretreated sample after enzymatic hydrolysis. An enzyme activity rate was 70 FPU/mL and 40 CBU/mL, and loading time was 24, 48, 72 h. The monosaccharide in pretreatment sample after enzymatic hydrolysis was analyzed by high performance liquid chromatography (HPLC). The changes of physico-chemical properties were determined by fourier transform infrared spectroscopy (FTIR) and X-ray diffractometer (XRD). FTIR was investigated the chemical changes of the kenaf core due to pretreatment. The structural changes of cellulose due to chemical reaction induced by gamma irradiation were investigated using XRD. |
