The specific binding interaction between poly(3-hydroxybutyrate) and the substrate binding domain of poly(3-hydroxybutyrate) depolymerase probed by two-dimensional infrared correlation spectroscopy |

We examined the specific binding interaction between the substrate binding domain (SBD) of poly(3-hydroxybutyrate) (PHB) depolymerase and PHB using two-dimensional infrared correlation spectroscopy. Some of the amino acid residues in the SBD are expected to contribute to the specific biding of the SBD on the surface of PHB. Positively charged histidine and arginine residues seem to have electrostatic interaction with the carbonyl group of PHB, and asparagines and glutamine with polar side chains are also suggested to be involved in the molecular recognition between the SBD and PHB. We measured the infrared spectra of the PHB film in the presence of SBD solution at different humidity. The cross-peaks in the 2D synchronous correlation spectra indicate that the amino acid residues with hydrophilic side chains have interactions with carbonyl groups in the PHB. |
