제목 Gold Quenched Near-Infrared Fluorescence Imaging Probe
초록 The optical imaging probe for cancers was successfully developed based on NIRF fluorophore (Cy5.5) labeled MMP-2 substrate on the surface of gold nanoparticles. Super quenching effect of Cy5.5 fluorescence on the surface of gold nanoparticles was gained by fluorescence resonance energy transfer between closely located Cy5.5 fluorophores as well as by surface resonance plasmon effect of gold nanoparticles. After the activation by the presence of MMP-2, however, the peptide connection between gold nanoparticles and Cy5.5 was selectively cleaved and Cy5.5 became fluorescent. The probe recovered strong fluorescent signals after the enzymatic degradation in vitro and in vivo. Considering all the fact, the proposed optical probes are potentially excellent candidates for the field of enzyme activity assay in vitro and in vivo imaging of tumors.
