Nano-Sized Ionic Complex Based on Insulin and Glycol Chitosan as a Potential Therapeutics for Type I Diabetes |

Glycol chitosan (GC) and its derivative bearing a hydrophobic moiety were investigated as the potential carrier for the protein drug, insulin. Both of GC and its derivative could form stable nano-sized complexes with insulin via the ionic interactions. The complex had a size in the range of 100-500 nm, depending on the weight ratio of the polymer and insulin. The biological activity of the insulin in the complex was comparable to that of native insulin, indicating that the complex formation does not affect the biological activity of insulin. From in vivo results, it was demonstrated that, when subcutaneously injected into the streptozotocin-induced type I diabetic mice, the nanocomplexes effectively decreased the blood glucose level for the longer period of time than native insulin. Overall, the nanocomplexes, prepared in this study, might be useful as the therapeutics for type I diabetes. |
