In this study, we introduced polyamidoamine (PAMAM) generation 4 to conjugate a cell penetrating peptides (CPPs) sequence (KFF, FFK) and transform KFF into FFK in order to develop an outstanding nano carrier. Cell penetrating peptides are well known for being effective delivery carriers into the cell membrane, and CPPs is an approximately short peptide sequence which is originated from PTD (Protein-Transduction Domain) or MTS (Membrane-Trans locating Sequence). Generally, it has a different cellular uptake into the cell membrane as CPPs doesn't damage the cell membrane as it is delivered to the internal part of the cell. Although DNA or macro molecule can't get through, that substance can be delivered into cell though. For this reason, we conjugated cell penetrating peptides with a surface of dendrimers. By introducing lysine, we expect to confirm the enhanced transfection efficiency, Cytotoxicity compared with PAMAM G4 in various cell lines. |
