In order for nonviral vectors to be clinically useful, it will be desirable to develop formulations that mediate high levels of cell-specific gene expression with minimal toxicity profiles. Here we propose a highly efficient polyethylenimine (PEI)-based non-viral vector that facilitate intracellular reducible environment triggered gene release, low cytotoxicity and angiogenic endothelium homing characteristics. We developed bioreducible polymer synthesized by cross-linking of low molecular weight thiolated PEI via potentially biodegradable linkages, which showed enhancement in gene delivery efficiency with minimal cytotoxicity. Especially, the bioreducible gene carriers are functionalized on their surface with novel cNGR (Asn-Gly-Arg) peptide using easy one-pot modification chemistry. In this report, the synthesis of gene carriers, characterization, and transfection efficiency will be studied in conjunction with their tumor-targeting ability. |
