In this study, a novel cationic dendrimer was synthesized consisting of poly(amidoamine) dendrimer (PAMAM, generation 4) backbone with both L-arginine (Arg) at termini and 6-aminohexanoic acids (Ahx) between the original core polymer and peripheral Arg units. The synergistic chemical modification of PAMAM with Arg and Ahx at the same time resulted in higher transfection efficiency with much less cytotoxicity. From the gene transfection experiments, PAMAM-Ahx-Arg showed much higher transfection ability compared to PAMAM or PAMAM-Ahx. Furthermore, it was found that PAMAM-Ahx-Arg showed very low cytotoxicity compared with PEI25KD and even with PAMAM-Arg. It can be confirmed that the Arg-grafting of PAMAM dendrimer can endow a higher capability for the transfection as noted in the previous study, while the Ahx spacer increases hydrophobicity to the dendrimer introducing the flexibility of the conjugated amino acids and reduces cytotoxicity. |
