In this study, we show that firefly luciferase (LUC) induces in vitro amyloid polymorphism of the Parkinson’s disease-related protein of α-synuclein by acting as a novel template. The drastically accelerated fibrillation kinetics of α-synuclein with LUC indicates that the seed formation has required the biochemical activity. In addition, fluorescent dye binding, electron microscopy, and Fourier transformed infrared spectroscopy revealed the morphological differences of the α-synuclein amyloid fibrils prepared in the absence or presence of LUC. As the altered morphological characteristics became inherent to the matured fibrils, those properties were inherited to next-generations via nucleation-dependent fibrillation process. Therefore, the seed control by addition of LUC would be an effective means to earmark amyloid fibrils with different biochemical characteristics. |
