Highly Fluorescent Graphene Oxide Biosensors for Determination of AChE Activity Involved in Alzheimer`s Disease |

Alzheimer`s disease is caused by the self-assembling of amyloid beta peptides producing fibril and their accumulation in the brain, which is known to be accelerated by acetylcholinesterase (AChE) catalyzing the hydrolysis of acetylcholine (ACh) to give choline and acetic acid. Herein, we demonstrate a simple approach for designing a graphene oxide (GO) fluorescence-based biosensor for the detection of AChE activity and inhibitors. GO was non-covalently functionalized with phenoxy-modified dextran polymer (PhO-dex-GO), leading to significant improvement of its colloidal and optical stability in various media solutions. The fluorescence of PhO-dex-GO increased as acetylthiocholine (ATCh) was hydrolyzed by AChE, but no fluorescence increase was observed without the enzymatic hydrolysis reaction. Based on this turn-on fluorescence response, the PhO-dex-GO sensor was able to report kinetic and thermodynamic parameters in relation to AChE activity and inhibition. |
