Intervertebral disc (IVD) degeneration is associated with increased cell senescence and apoptosis. For the 3D culture of tissue engineering, hydrogel and mesh structures are widely used. Using electrospinning methods, we fabricated PCL nanofiber for mesh structures and using alginate, hydrogel was fabricated. For the cellular approaches of disc tissue engineering, human mesenchymal stem cells (hMSCs) provide plausible cell source and cell co-culture system of hMSCs and IVD cells is proved for notable efficacy in cell differentiation. We first seeded hMSCs onto PCL nanofibers. After attachment, IVD cells were seeded into alginate hydrogel which encapsulating PCL nanofibers. Each hMSCs and IVD cells were seeded on PCL nanofibers and in alginate hydrogel as control. Our study suggests that the hMSCs can be differentiated into discogenic phenotype by co-culture, which provides excellent mechanism for hMSCs based tissue engineering for IVD regeneration. |
