In vivo delivery of anti-KITENIN shRNA complexed with biodegradable PEI-alt-PEG suppressed colon carcinorma established subcutaneously in mice |
초록 |
Non-viral gene therapy including polymeric carrier has gained increasing interest as an alternative to viral vectors. In this study, biodegradable gene carrier was synthesized and explored for anti-cancer gene therapy in vitro and in vivo. It has been suggested that specific knockdown of KITENIN expression in colon adenocarcinoma leads to suppression of progression and metastasis of tumor in mice and can be achieved by the delivery of siRNA or shRNA targeting KITENIN. Therefore, Biodegradable gene carrier-mediated delivery of anti-KITENIN shRNA has been developed for efficient cancer gene therapy. In vivo systemic delivery of nanoparticles demonstrated the drastic suppressions of tumor growth by PEI-PEG over time. Our on-going study suggested that incorporation of tumor-specific ligands would improve its efficacy greatly. |
