제목 H2O2 responsive antithrombotic polymeric nanoplatforms
초록 Thrombosis is a meshwork of platelet and fibrin. Hydrogen peroxide (H2O2) is an important factor of platelet activation and induces inflammation. By taking advantages of H2O2 overexpression and fibrin, we designed blood clot targeting multifunctional nanoplatform (TTIAN), which was composed of antioxidant polymer (BAP-IR) and fibrin targeting lipopeptide (DSPE-PEG-CREKA). BAPIR was made up of H2O2 sensitive boronate, natural antioxidant Hydroxybenzyl alcohol (HBA) and near infrared dye (IR 820). TTIAN has thermodynamically size ~120 nm and has excellent biocompatibility and cell protection ability. The micelles have fibrin targeting activity and diminish the generation H2O2 in vitro. The ferric chloride induced thrombosis model on mice, the micelles could targeting thrombi, and reduce the vascular inflammation. Additionally tirofiban loaded micelles more effective anti-thrombotic effect than free drug injection.
