We report mitochondria-targeted photothermal nanoparticles, decorated with energy blocker 3-bromopyruvate (3-BP) and delocalized lipophilic triphenylphosphonium (TPP) cations to target the mitochondrial membrane of cell. Using catechol composited polymer, the carbonized fluorescent nanoparticles [FNPs(C-PgD)] were efficiently combined with polyethylene glycol (PEG) grafted poly (dimethyl aminoethyl methacrylate)-q-boronic acid (PEG-g-PDMA-BA) by diols cross linking reaction which, were also conjugated with IR825, TPP and 3-BP. In vitro studies confirmed a profound anticancer activity of TPP/BA-PgD@FNPs (C-PgD) when, comparing between cancer and normal growing cells line, as well as in absence and presence of near infrared (NIR) light treatments. We thereafter expected that blocking mitochondria metabolic activity in a photothermal based therapeutic system might open a new pathway for cancer therapy. |
