제목 Comparison of chemical and physical cross-linked three-dimensional gelatin scaffold
초록 The bioink is one of the most important factors in three-dimensional (3D) bioprinting. Especially, natural polymers such as gelatin having high biocompatibility are attractive materials. However, gelatin has poor mechanical properties and cross-linking is required to maintain the 3D structure. N-Hydroxysuccinimide (NHS) is widely used for gelatin cross-linking, but residual NHS can be toxic to cells. In this study, physical cross-linking via dried heat treatment (DHT) was used to reduce cytotoxicity caused by chemical cross-linking. We fabricated 3D scaffold using 3D bioprinting and cross-linked by DHT. We used a SEM to determine the surface morphology of 3D scaffold and chemically bond exchange was confirmed by FT-IR. For evaluating the biological effect on 3D scaffold cross-linked by DHT, human dermal fibroblast was cultured on 3D scaffold. The DNA assay shows that the cell proliferation of DHT scaffold was 44% higher than NHS cross-linked 3D scaffold after 14 days of culture.
