제목 Molecularly Imprinted Polymers Having Amidine and Imidazole Funtionalities for Enzyme-mimetic Esterolytic Catalysis. II
초록 A molecularly imprinted polymer (MIP) having both amidine and imidazole functional groups in the active sites has been prepared using p-nitrophenyl phosphate as a transition state analogue (TSA). The ability of the imprinted polymer to imitate enzymes in ester hydrolysis has been evaluated. The rate enhancement of the imprinted polymer was 60 folds over the uncatalyzed reaction in background buffer solution and two folds compared with the control blank polymer having both functional groups but prepared in the absence of the TSA template. Possible cooperative effect between amidine and imidazole is proposed in hydrolysis of p-nitrophenyl methyl carbonate (NPMC) as a substrate when both groups were arranged in proximity by molecular imprinting. The Michaelis-Menten kinetics of the hydrolysis with MIP was measured in buffer at pH 7.0 with p-nitrophenyl acetate as a substrate. Km and kcat were calculated to be 1.06 mM and 0.137 h-1, respectively.
