Eggshell Membrane Sandwich for Bone Tissue Regeneration |

Eggshell membrane (ESM) is a highly collagenized fibrous double-layered sheet, located between the calcified eggshell (outer membrane, OM) and the egg white (inner membrane, IM), which also serves as the scaffold for the formation of the eggshell. In this study, adipose-derived stem cells (ADSC) were sandwiched between the double layers of ESM isolated form chicken eggs were investigated for bone tissue engineering. The inner and outer membranes were evaluated for their individual morphologies using SEM and contact angle apparatus then tested individually and in combination for tensile strength. Apatite layer formation on the membrane surfaces were also tested using simulated body fluids. Since the OM showed more apatite formation, two layers of OM were used to sandwich ADSC and the semi-permeability of the membranes still allowed cell proliferation and differentiation in vitro. The sandwich of OM-ADSC-OM was also implanted on critical sized cranial defects of male Sprague Dawley rats for 30 days then extracted. Micro-CT analysis and histological staining using hematoxylin and eosin staining as well as Masson’s Trichrome staining methods showed significant bone formation. Thus, eggshell membrane can serve an application on bone tissue regeneration. |
