제목 A MALDI-MS-based quantitative glycoprofiling method on a 96-well plate platform
초록 Abnormal glycation changes are closely related to various human diseases, and the glycation profile has been used as a biomarker for the diagnosis of various diseases. Changes in protein glycosylation also have a direct effect on protein structure and function. In this study, we developed a high-throughput MALDI-MS based quantitative targeted glycomics (HT MALDI-QTaG) method for total N-glycan analysis. Despite the chemical derivatization process performed in 96-wells with built-in 10 kDa MWCO membrane filter, MALDI-MS showed excellent quantitative linearity (R2>0.99) between the various amounts of target glycoprotein and MALDI peak intensity. We also confirmed relative quantitative reproducibility at different well locations in 96-well plates. In addition, the HT MALDI-QTaG method proposed in this study was successfully used for the analysis of bovine fetuin, human serum and Enbrel®.
