Cytocompatible self-assembling hydrogel and its modular modification using highly selective host-guest chemistry |

Self-assembling hyaluronic acid (HA) hydrogel (CB[6]/DAH-HA hydrogel) was successfully developed using highly specific host-guest chemistry with cucurbit[6]uril-modified HA (CB[6]-HA) and 1, 6-diaminohexane-modified HA (DAH-HA). CB[6]/DAH-HA hydrogel could be easily decorated with various functional groups by facile and modular modifications maintaining the structural integrity of the hydrogels. According to live/dead cell assay with calcein AM and ethidium homodimer-1, more than 90% of NIH3T3 (mouse fibroblast) cells in the hydrogels emitted the green fluorescence of live signal even after encapsulation for 3 days. Moreover, the facile modular modification of CB[6]/DAH-HA hydrogel encapsulating NIH3T3 cells with cyclic(RGDyK)-CB[6] resulted in dramatically enhanced cell adhesion and proliferation. The novel self-assembling cytocompatible CB[6]/DAH-HA hydrogel system would be exploited as in situ forming hydrogels for various cell therapy and tissue engineering applications. |
