Cytochrome P450 (P450) is one of monooxygenase which contains heme as a cofactor. It is widely found in eukaryotes and several prokaryotes also contains P450. It is known to be responsible for biosynthesis of secondary metabolites or detoxification of external compounds within cell. Among P450 families, there are self-sufficient P450 groups, fusion proteins of P450 and reductase. Compared to other P450 families, P450 in such families show apparently high turnover number. Widely progressed genome sequencing studies have found Streptomyces species, well-known for various secondary metabolites, contain various P450 genes, many involved in biosynthesis of natural antibiotics. However, there are still lots of P450 to be characterized. In this study, one of self-sufficient P450 from Streptomyces cattleya, assigned in CYP102 family, was cloned and expressed in Escherichia coli. |
