Cell culture is an important technique that manipulates cells to study about several physiological and biological parameters. Among various cell culture methods, 3D cell culture has emerged to study complex metabolism with a close duplication of the natural in vivo environment. However, 3D cell culture has several disadvantages associated with the 2D culture system. The 3D cell cultures also used flat plate to offer stiff platform for 3D constructs. As consequence, the cells in 3D constructs spread and unnatural grow on the surface of plate. Therefore, the natural microenvironment and the phenotype of the cells is not duplicated in vitro. In this study, we overcome problem of cell adhesion between 3D construct and culture plate using hexanoyl glycol chitosan which have ultra-low cell binding characteristic. This system accomplished higher growth, realistic morphology, and homogeneous cell distribution that failed to obtain in a 3D culture on plain plate. |
