Cyclic β-1,2-glucans, cyclosophoraoses (Cys), synthesized in the family Rhizobiaceae, have been used various biotechnological applications by forming complexes with target molecules. In this study, we isolated and characterized novel anionic Cys using combined chromatographic techniques such as P-6, P-2 gel permeation, DEAE-cellulose anion exchange chromatography. The degree of polymerization of anionic Cys was investigated by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The precise chemical structures of novel anionic Cys were elucidated by various NMR spectroscopic analyses, including 1H, 13C, 31P NMR and 2D HSQC NMR. Various umber of succinyl and phosphoglycerol substituents at the C6 position of Cys were identified by FT-IR spectroscopy and MALDI-TOF-TOF MS. Target ligand complexation studies using isolated novel anionic Cys will be utilized at drug delivery system or chiral technology. |
