To overcome the threat of cancer, chemotherapeutics are widely utilized to control and eliminate the tumor. Although they are proven to be an efficient method in cancer therapy, traditional chemotherapeutic agents induce various side effects against the normal cells due to the lack of delivery systems. Therefore to enhance therapeutic efficacy towards cancer, design of the functional carrier system is essential. Owing to the achievement in biochemical science and supramolecular chemistry, nano sized carriers with diverse functionality are being developed. Herein, cyclodextrin (CD) polyrotaxane (PR) structure was synthesized as a targeted and stimuli responsive drug delivery carrier. A DNA aptamer, i-motif and its complimentary DNA were each conjugated to αCD. With polyethyleneglycol, it formulates a PR based nanoconstruct (NC). Because of its size and aptamer, NC is targeted to cancer cells both passively and actively. It releases drug in pH responsive manner after endosomal uptake. |
